  method

  After    anhydrous ethanol-induced gastric mucosal injury in rats, polyprezinc 100mg/kg, 200mg/kg and excipients were administered respectively. The blank group was used as a control to detect the expression of HSP70 protein in the gastric mucosa. At the same time, the gastric mucosa IGF-1 content and SOD activity of rats in each experimental group were detected.

  Result

  The relative gray value of HSP70 expression in the polyprezinc 100mg/kg treatment group was 278.3% ± 10.8% at the 3rd day of treatment; the relative gray value of HSP70 expression in the polyprezine 200 mg/kg treatment group was 471.1% ± 24.7 at the 3rd day of treatment %, compared with the blank group and the excipient group, the difference was statistically significant (P<0.01). There was no significant difference in the content of IGF-1 and SOD activity in the gastric mucosa of rats in each experimental group (P>0.05).

  in conclusion

  Polyprezinc can induce large amounts of HSP70 in the gastric mucosa of rats after injury, and play a role in protecting the gastric mucosa.

  Foreword

  Heat shock protein 70 (HSP70) is one of the main proteins in the heat shock protein family. HSP70 exists in the gastric mucosa of humans and animals. The gastric mucosa will quickly synthesize HSP70 after being stimulated by external sources (alcohol), which has a protective effect in the process of acute gastric mucosal injury. Relaisen (Polyprezinc Granules) is a chelate of L-carnosine and zinc, which can induce the production of heat shock protein, thereby exerting an anti-damage effect.

  1 Materials and methods 1.1 Animals

  SD rats, purchased from the Laboratory Animal Science Department of Peking University Health Science Center, license number: SCXL (京) 2006-0008.

  1.2 Drugs

  Polyprezinc bulk drug powder, provided by Jilin Province Boda Weiye Pharmaceutical Co., Ltd. The polyprezinc powder was dissolved in a 1.5% carboxymethyl cellulose solution (CMC), and the concentration of 100mg/kg and 200mg/kg drugs were prepared for use.

  1.3 Method 1.3.1 Experimental animals

  20 male SD rats, weighing 200-250g, were raised in standard environmental temperature (22°C ± 2°C) and humidity (55% ± 5%), and a standard animal laboratory with alternating light and dark cycles of 12 hours, free to eat and drink. And start the experiment 1 week after the animals adapt to the environment.

  1.3.2 Experiment grouping

  20 rats were randomly divided into 4 groups, 5 in each group. 5 rats in the blank control group: free diet, 5 rats were sacrificed on the 3rd day, and the rats were fasted for 24 hours before being killed. There were 5 animals in each group in the experimental group. All animals were fasted 24 hours before the experiment and could not help water; CMC group: 1ml absolute ethanol was given by gavage, 1 hour later, 1ml 1.5% CMC was given by gavage, and then every day at the same time for 2 consecutive days. Animals were sacrificed 1h after the last administration; the polyprezine 100mg/kg treatment group: 1ml of absolute ethanol was given by intragastric administration, 1 ml of polyprezine 100mg/kg was given by intragastric administration after 1h, and the same time every day thereafter for 2 days, and at the end Animals were sacrificed 1h after one administration; the polyprezinc 200mg/kg treatment group: 1ml of absolute ethanol was given by gavage, 1 ml of polyprezine 200mg/kg was given by gavage after 1h, and then every day at the same time for 2 days, and the last time Animals were sacrificed 1h after administration. The animals were put to death by intraperitoneal injection of pentobarbital sodium anesthesia.

  1.3.3 Sample collection

  After the rats of each group were sacrificed, the whole stomach was taken out by open surgery. Cut the appetizer along the greater curvature of the stomach, immediately spread it on a glass plate placed on ice, observe the changes in the gastric mucosa with naked eyes, and take pictures. Subsequently, the gastric tissue was taken, fixed in 10% formaldehyde solution for 6 hours, embedded in paraffin, sectioned, and stained with HE. Use clean glass slides to quickly scrape the gastric mucosa, store them in cryopreservation tubes and store them in liquid nitrogen, and store them in a refrigerator at -80℃ for long-term storage.

  1.3.4 Western blotting to detect the expression of HSP70 protein in rat gastric mucosa

  Place the 1.5ml centrifuge tube containing the gastric mucosa on ice, weigh the gastric mucosal tissue and add the lysate in proportion, cut the tissue, and homogenize with a homogenizer at 15000r/min. When homogenizing, the EP tube needs to be immersed in a mixture of ice and water for cooling. After the homogenization is completed, incubate on ice for 20min, centrifuge at 4℃, 13000r/min, 20min. After centrifugation, take the supernatant, aliquot and store for testing. The target protein was electrophoresed on a 10% SDS-PAGE gel and transferred to the membrane. Incubate with anti-HSP70 (1:1000 dilution) antibody overnight. The secondary antibody goat anti-rabbit IgG (diluted 1:10000) was incubated, washed, and exposed. Use software Labworks 4.6 to analyze the results.

  1.3.5 Enzyme-linked immunoassay (ELISA) to detect rat gastric mucosa insulin-like growth factor-1 (IGF-1)

  During the process of anesthesia, the rats were quickly taken blood from the heart and centrifuged; IGF-1 ELISA kit was used to detect the content of IGF-1 in the serum.

  1.3.6 Enzyme-linked immunoassay (ELISA) method for detection of superoxide dismutase (SOD) activity in rat gastric mucosa

  The gastric mucosa of rats was scraped by the above method, homogenized in Tris-HCl buffer (pH=7.4), centrifuged at 4°C, 10000r/min, 30min, and the supernatant was taken; SOD detection kit was used to detect SOD activity.

  1.4 Statistical analysis

  Use SPSS 11.5 statistical software for statistics. Data are expressed as mean±standard deviation, analyzed by One-Way ANOVA, and used Bonferroni test for multiple comparisons. P<0.05 indicates that the difference is statistically significant.

  2 Results and analysis

  2.1 Gross and pathological manifestations of gastric mucosa in each experimental group

  Compared with the blank control group, after intragastric administration of 1ml of absolute ethanol, polyprezinc 100mg/kg 1ml was given for 3 days, the gastric mucosa recovered better in general and under microscope. After intragastric administration of 1ml of absolute ethanol, 200mg/kg 1ml of polyprezinc was administered intragastrically for 3 days, and the gastric mucosa and pathology recovered well. However, the gastric mucosa of rats given CMC for 3 days did not change significantly (Figure 1).

  Using Western blotting method, the relative gray values ​​of HSP70 expression in the gastric mucosa of rats in the polyprezinc 100mg/kg treatment group and 200mg/kg treatment group were 278.3% ± 10.8%, 471.1% ± 24.7%, and blank Compared with the CMC group (100%) and the CMC group (153.3% ± 3.2%), the difference was statistically significant (P<0.01, Figure 2-3).

  2.3 Serum IGF-1 content in each experimental group

  The serum IGF-1 levels in the blank control group, CMC group, polyprezinc 100mg/kg treatment group and polyprezinc 200mg/kg treatment group were (1.35 ± 0.093) ng/ml and (1.269 ± 0.372) ng/ml, respectively , (1.489 ± 0.159) ng/ml and (1.427 ± 0.142) ng/ml, there was no significant difference in IGF-1 content between the groups (P>0.05).

  2.4 SOD activity of gastric mucosa in each experimental group

  The gastric mucosal SOD activity of the blank control group, CMC group, polyprezinc 100mg/kg treatment group and polyprezinc 200mg/kg treatment group were (48.68 ± 7.80) U/ml, (46.52 ± 2.79) U/ml, (57.04 ± 6.52) U/ml and (55.97 ± 8.00) U/ml, there was no significant difference in SOD activity between the groups (P>0.05).

  Discuss

  The study showed that the protein expression of HSP70 in the CMC group was slightly higher than that of the blank control group 3 days after modeling, indicating that HSP70 produced by stress exerted a protective effect after modeling. Reliance (polyprezinc particles) was applied to the injured gastric mucosa in rats, and the protein expression of HSP70 was significantly higher than that of the CMC group. That is, Reliance induced a large amount of HSP70 to produce, thereby accelerating the repair of gastric mucosal damage. Although HSP70 was also produced in the CMC group, it was not enough to fight the injury. Reliance continuously induced the production of HSP70 is the key to fight the injury. At the same time, studies have shown that IGF-1 and SOD also play an important role in gastric mucosal injury. The data in this experiment show that the IGF-1 level in the Relaisen treatment group has a rising trend, and the SOD level is slightly higher than that in the untreated group. In summary, Relaisen (Polyprezinc Granules) is a new gastric mucosal protective agent with dual mechanisms of protection and repair. It can induce the damaged mucosa to produce a large amount of HSP70, increase the level of IGF-1 and SOD, thereby Play the role of protecting and repairing gastric mucosa.
