Urease has a complex structure, with a thickness of about 13nm and a regular triangle symmetry. It is a nickel metalloenzyme, which is determined by the nickel content of the active site. The urease operon contains 7 genes, namely ureABIEFGH. Among them, ure A and ure B are structural genes, ure EFGH is an accessory gene, and ure I is a gene specific to Hp urease. The urease accessory gene can assist the insertion of nickel into the active site of the UreA/ UreB protein to form an active protein. The activation of the enzyme activity of urease requires the insertion of two nickel in the six active sites of the holoenzyme.
