  Preface

  Healthy killer—oxygen free radicals are rich in the air we live in. We can't survive without oxygen, but oxygen also has a harmful side to the human body. Sometimes it can kill the healthy cells of the human body. Of course, it is not oxygen itself that directly kills cells, but a kind of harmful substance called oxygen free radicals produced by it. It is a metabolite of the human body, which can cause damage to the biofilm system and cause great harm to human health.

  In mammalian cells, there are free radical scavenging enzymes, such as superoxide dismutase (SOD), which can scavenge superoxide free radicals (O2¯), glutathione peroxidase (GSH-px), and it Can remove hydrogen peroxide (H2O2) generated by SOD. If the production of oxygen free radicals in the gastric mucosa exceeds the ability of free radical scavenging enzymes to scavenge free radicals, then the gastric mucosa may be damaged by free radicals.

  Polyprezinc is a chelate composed of zinc and L-carnosine, which is clinically used to treat gastric ulcers. Studies have shown that polyprezinc can inhibit the production of superoxide by neutrophils and has an antioxidant effect in vitro. Is it difficult, can polyprezinc affect the synthesis or activity of gastric mucosal free radical scavenging enzymes to protect the gastric mucosa? Please see the following research.

  Materials and Method

  1.1 Animal

  Male SD rats, weighing 170-190g, fasted for 24 hours before the experiment and reared in separate cages.

  1.2 Evaluation of gastric mucosal injury

  1 hour before the rats were given ethanol, use 1 mL/100g body weight to accumulate Prezinc (1, 3, 10, 25 mg/kg), ZnSO4·7H2O (10, 25 mg/kg/day) and L-carnosine ( 7.7, 19 mg/kg) pretreatment. The rats were intravenously injected with human Cu and Zn-SOD (15,000, 30,000, and 45,000 U/kg) at a volume of 0.5 mL/100 g body weight 10 minutes before giving ethanol. One hour before necrotic agent pretreatment, allopurinol (30 and 100 mg/kg, 1 mL/100g body weight) was intraperitoneally injected. 0.5% methyl cellulose and 0.9% NaC were used as vehicle controls. After intragastric administration of 1 mL of 100% ethanol for 1 hour, the rats were anesthetized with ether, and the stomach was cut along the greater curvature. Measure the length (mm) and width (mm) of the red band of the gastric mucosa by a stereo microscope, and calculate the band area (mm²) to evaluate the degree of gastric mucosal damage. The total area of ​​the band is expressed by the lesion index.

  1.3 SOD activity

  After pretreatment with polyprezinc (10 mg/kg) for 0, 0.5, 1, 2, 3, and 5 h, the rats were sacrificed. The stomach was cut along the greater curvature of the stomach, and the gastric mucosa was scraped. The upper part was determined by the nitrite method. SOD activity in the clear liquid.

  1.4 SOD and GSH-px activity

  Pretreatment with polyprezinc (3, 10, 25 mg/kg), ZnSO4·7H2O (10, 25 mg/kg/day) and L-carnosine (7.7, 19 mg/kg) 1 hour before intragastric administration of 60% ethanol , The control group was pretreated with 0.5% methylcellulose, and 1 hour later, 1 mL of 60% ethanol was given to the rats. After 1 hour, the rats were sacrificed and the stomach was taken. The SOD in the rat gastric mucosa was determined by the nitrite method, Paglia method and Valentine method. Activity and GSH-px activity.

  Pre-treatment with polyprezine (10 mg/kg) 1 hour before intragastric administration of 60% ethanol, 30 min before intragastric administration of intragastric ethanol (60% 1mL), immediately subcutaneous injection of cycloheximide (6 mg) /kg), with 0.5% methylcellulose (1mL/100g body weight) as a carrier, and observe the gastric mucosal injury and the activities of SOD and GSH-px after 1 hour.

  1.5 TBA active substance

  After gavage with 1 mL of 60% ethanol, the rats were sacrificed at 0, 5, 30 and 60 min, and the stomach was taken. The Ohkawa method was used to determine the TBA active substances in rat mucosal tissues as an indicator of lipid peroxidation.

  1.6 Statistical analysis

  The results are expressed as the mean ± S.E., using one-way analysis of variance to analyze the data, and Duncan multiple interval test to determine the statistical significance of the differences between the groups.

  Results and analysis

  2.1 The effect of polyprezinc on alcohol-induced gastric mucosal injury

  Pretreatment with polyprezinc (1-25 mg/kg) 1 hour before gavage with 100% ethanol can reduce the pathological index in a dose-dependent manner, indicating that polyprezinc can effectively prevent gastric mucosal injury, among which, 10 and 25 mg/kg The kg dose has the most significant effect. Allopurinol and human Cu and Zn-SOD can also protect the gastric mucosa.

  2.2 Polyprezinc increases SOD activity in normal rat gastric mucosa After pretreatment with polyprezinc (10mg/kg), the gastric mucosal SOD activity was measured at different time points. The results showed that the SOD activity increased significantly at 0.5h until 2h. In addition, the SOD activity was significantly increased by 33-42% after pretreatment with polyprezinc (3-25mg/kg) for 1h.

  2.3 Polyprezinc increases the activity of SOD and GSH-px in gastric mucosal injury induced by 60% ethanol

  One hour after    gavage with 60% ethanol, the SOD activity of rats in the control group was significantly higher than that in the normal group (36.2%). Polyprezinc (10 and 25mg/kg) was pretreated 1 hour before intragastric administration with 60% ethanol. Compared with the control group, the activity was significantly increased by 58% and 42%, respectively.

  When the GSH-px activity in rats was measured under the same conditions as the SOD activity, the GSH-px activity in the control group was significantly higher than that in the normal group (34.4%). Compared with the control group, after pretreatment with polyprezinc (10 and 25 mg/kg), the activity of GSH-px was significantly increased by 39% and 40%.

  2.4 The effect of polyprezinc combined with cycloheximide on the activity of SOD and GSH-px in gastric mucosal injury induced by 60% ethanol

  The gastric mucosal damage induced by 60% ethanol is less severe than that of 100% ethanol (injury index: 60% ethanol, 16.0±6.2mm², 100% ethanol, 97.1±11.7mm²) (Figure 4). Polyprezinc (10mg/kg) can significantly prevent mucosal damage induced by 60% ethanol, with a prevention rate of 88%. In addition, polyprezinc improves SOD activity by 58% and GSH-px activity by 39%. However, treatment with cycloheximide (6mg/kg×2) completely eliminated the preventive effect of polyprezinc (10mg/kg) on ​​mucosal damage and the increase in SOD and GSH-p X activity.

  2.5 The effect of polyprezinc on TBA-responsive substances in gastric mucosal injury induced by 60% ethanol

  5min after intragastric administration of 60% ethanol, the active substance of TBA increased significantly, and it was 80% higher at 60min than at 0min. Ethanol gavage for 60 minutes after the increase of TBA active substances, after pretreatment with polyprezinc (10 and 25mg/kg), the increase of TBA active substances was inhibited by about 50%.

  2.6 The effect of ZnSO4·7H2O and L-carnosine on SOD and GSH-px activity

  Neither ZnSO4·7H2O (10mg/kg) nor L-carnosine (7.7mg/kg) can effectively protect the gastric mucosa injury induced by 100% ethanol. After increasing the dose, only 25mg/kg of ZnSO4·7H2O can prevent the injury. In addition, It can also significantly increase the SOD activity of the gastric mucosa induced by 60% ethanol, and has a tendency to increase the activity of GSH-px.

  Discuss

  Polyprezinc can significantly increase the SOD and GSH-px activities of gastric mucosa that are damaged by 60% and 100% alcohol, but zinc ions (ZnSO4·7H2O) and L-carnosine alone cannot effectively protect the gastric mucosa induced by 100% ethanol. The damage indicates that the combination of zinc ions and L-carnosine in polyprezinc has a synergistic effect, thereby significantly improving the activity of oxygen free radical scavenging enzymes and achieving the effect of protecting the gastric mucosa.
