Experiment Overview
purpose
ICR mice were gavaged with 2% dextran sodium sulfate (DSS) to induce an animal model that can restore the course of UC Ulcerative colitis, and observe the therapeutic effect of polyprezinc on UC and its preliminary therapeutic mechanism.
method
ICR mice were randomly divided into the following 5 groups, each with 8 mice.
Experimental results
The general activity observation of experimental animals. During the experiment, in addition to the normal control group, the experimental group mice have different degrees of weight loss and activity reduction after DSS administration. The mice in the normal control group had normal activities. During the experiment, no mice in each group died.
On the 3rd day after the administration of DSS to mice, the weight of the mice increased slowly, and the weight difference compared with the normal control mice was very significant P<0.01. Polyprezinc enema administration had little effect on the weight loss caused by DSS. The weight recovery is not obvious. The experimental results are shown in Table 1, Figure 1.
DSS gave mice diarrhea after intragastric administration. Polyprezinc enema administration has an inhibitory effect on DSS-induced diarrhea in mice. There is a significant difference between the 120 mg/kg body weight treatment group and DSS model group. P<0.05, diarrhea was mild.
Polyprezinc can reduce the increase of inflammatory factors TNF-α and NF-κB in an animal model of UC Ulcerative colitis (UC Ulcerative colitis) caused by DSS, with statistically significant differences. Polyprezinc can increase the reduction of HSP in animal models of ulcerative colitis caused by DSS, and can inhibit the phosphorylation of AKT in the inflammatory signaling pathway.
Polyprezinc affects the blood picture of an animal model of ulcerative colitis caused by DSS. The symptoms and pathological manifestations of UC in mice caused by DSS are similar to those of human UC, and inflammation accompanied by increased peripheral blood leukocytes and neutrophils is similar to human UC.
The histopathological observation of the DSS-induced ulcerative colitis model of Polyprezinc showed that the colonic villi of the DSS group were arranged irregularly, the inflammatory cells increased significantly, and the mucosa was damaged. The pathological observation of the polyprezinc treatment group under the light microscope showed that the villi were intact and the villi showed a tendency to recover.
